Semaglutide is a synthetic peptide derived from the naturally occurring glucagon-like peptide-1 (GLP-1).1,2 GLP-1 is an incretin hormone, naturally secreted in the gastrointestinal tract, that stimulates insulin and suppresses glucagon secretion depending on glucose levels.1,3 As a GLP-1 receptor agonist, semaglutide was developed based on the body of knowledge gained from the development of liraglutide, but designed to have a longer half-life.4 The structural differences between GLP-1 and semaglutide reduce enzymatic degradation and promote albumin-binding, both of which increase the half-life of the peptide.5,6
